Glypican-3mRNA在人原发性肝细胞癌组织中的表达及意义

摘　　要：目的:探讨Glypican-3mRNA在人肝细胞癌中的表达和意义。方法:收集48例肝细胞癌肿瘤组织,相应39例癌旁组织和31例非肿瘤肝组织,采用RT-PCR检测Glypican-3mRNA的表达。结果:48例肝细胞癌肿瘤组织中有37例(77.1%)检测到Glypican-3mRNA表达,其中过度表达率为62.16%。而在相应的癌旁组织和非肿瘤肝组织中表达缺失。肝细胞癌Glypican-3mRNA表达与其瘤体大小和病理分级之间有显著性差异(P<0.05),而与患者年龄、性别、AFP值、HBsAg及包膜状况之间无显著性差异(P>0.05)。结论:Glypican-3mRNA在人肝细胞癌肿瘤组织中过度表达,可作为诊断肝细胞癌的肿瘤特异性标记物之一。Objective： To investigate glypican-3 mRNA expression and its significance in human hepatocellular carcinoma. Methods： The expressions of glypican-3 mRNA were detected by reverse transcriptase polymerase chain reaction （RT-PCR） in 48 cases of hepatocellular carcinoma tissues, 39 para-cancer tissues and 31 normal hepatocellular tissues respectively. Results： Overexpression of glypican-3 mRNA was found in 37/48 （77.1% ） hepatocellular carcinoma tissues. But glypican-3 mRNA was silence in both paracancer tissues and normal hepatocellular tissues. Significant relationship was observed between positive expression of glypican-3 mRNA and tumor body size and pathology grading in hepatocellular carcinoma （P〈 0.05）. There were no difference between Glypican-3 mRNA express and patient age, sex, AFP value, HBsAg and integument state （P〉0.05）. Conclusion： The overexpression of glypican-3 mRNA in hepatocellular carcinoma may be a new gene diagnosis index of hepatocellular carcinoma.
