摘 要:应用流式细胞仪及细胞活性染料Dio,PI对高钾角膜保存液保存后角膜内皮细胞的存活与K液进行了对照实验。结果表明:(1)高钾保存液组保存5、7、9、11天的角膜内皮细胞存活率均高于K液组(P<0.05);(2)含钾离子浓度为12.15mM的高钾保存液角膜内皮细胞存活高于共它高钾组(P<0.05)。提示低温状态下高钾角膜保存液对延长角膜内皮细胞的存活有一定的作用。A dual-color flow cytometric assay (two dyes: cyanine membrane dye-3, 3'-Dioctadecyloxaearboeyanine perehlorate and propidium iodide) was used for the assessment of bovine corneal endothelial cell survival in potassium-rich solution and K medium (prepared by Kaufman method) for 3, 5, 7, 9, and 11 days at 4℃. Results showed that (1) the rate of survivable endothelial cells stored in potassium-rich solution was significantly higher than that in K medium for 5, 7, 9, and 11 days respectively (P<0.05); (2) among the potassium-rich solutions with various K + concentrations used, the rate of survivable endothelial cells stored in the potassium-rich solution with K+ concentration of 12.15 mM was the highest. It is suggested that under low temperature, potassium-rich solution play a role in prolongation of corneal endothelial cell survival.
