摘 要:以研究外源基因有效发送至靶细胞并高效表达为目的,为了获得能提高外源基因在机体内表达量的增强剂,以pCMVβ-gal作为报告基因,分别辅以油酸和透明质酸酶两种不同的生化试剂,注射小鼠后肢腿部肌肉。通过检测在两种不同的生化试剂作用下,pCMVβ-gal在机体内表达量的差异,以期获得能提高外源基因表达量的增强剂。结果证明油酸和透明质酸酶均能有效提高pCMVβ-gal在小鼠体内的表达量,其中透明质酸酶引起的增强效果更为显著,是较好的基因表达增强剂。To achieve a higher efficiency with the exogenous gene expression is still a big challenge for the non-viral vector delivery approach in despite of the technology been improved over a decade. The main obstacle is to delivery the exogenous gene into tissues in vivo with a mimmal loss ann with a maximal penetration into cell membrane. Several strategies have been applied to achieve this goal previously, such as lipidsome or polymer encapsulations, gene gun, in situ electroporation. Chemicals with a permeable property might be considered as facilitators to enhance the egiciency of the exogenous gene entry. Owing to the easiness in application and cost effectiveness, two biochemical reagents have been incorporated to facilitate such delivery that may improve the exogenous gene expression in vivo. After injections of the mice intramuscularly with a pCMVβ-gal construct mixed with the oleic acid or hyaluronidase, it showed that the expression of the β-gal gene was enhanced by either of the facilitators compared with the mice injected with the β-gal gene formulated in saline. The enhanced expressions are apparently associated with the characteristics of the chemicals used since they are enable to increase the permealibility of ceils as previous documented. An evaluation of DNA vaccine formulated in the two facilitators is currently underway. Preliminary observations on the enhancement of the expressed exogenous gene using the two biochemical reagents may lead to the improvement for the development of DNA vaccination and gene therapy.
