摘 要:目的应用cDNA微阵列杂交技术筛选垂体瘤组织差异表达基因,并初步讨论了差异表达基因在垂体瘤发病过程中的作用。方法用cDNA微阵列技术筛选垂体瘤组织差异表达基因,对于在3种垂体瘤中高差异表达基因用RTPCR和实时定量PCR进行初步验证。结果用cDNA微阵列技术筛选垂体瘤组织差异表达基因按照基因功能分类,发现差异表达的基因主要与信号转导相关和发育、分化相关。PP4R2,GHRHR和clusterin分别在泌乳素瘤,生长激素瘤,无激素瘤中明显高表达,并经RTPCR,实时定量PCR等证实。结论PP4R2,GHRHR,clusterin可能分别在泌乳素瘤,生长激素瘤,无激素瘤的发生发展中发挥了重要作用。Purpose To screen differentially expressed gene profiles in pituitary adenomas,compare with that in normal pituitary, and to discuss the possible role of candidate genes in the pathogenesis of pituitary adenoma. Methods The differentially expressed genes in pituitary adenomas were studied by complementary DNA microarrays, and confirmed by RT-PCR and realtime-PCR, Results In comparison with that in normal pituitary, the genes involved in signal transduction pathway, growth and differentiation are mainly differentially expressed, PP4R2, GHRHR and clusterin are significantly highly expressed in prolactinoma, somatotroph and nonfunctioning adenomas respectively, which was also proved by RT-PCR and realtime-PCR, Conclusions PP4R2,GHRHR and clusterin may play a pivotal role in the development of prolactinoma,somatotroph and nonfunctioning adenomas respectively.
