摘  要:目的建立人肾癌裸小鼠原位移植转移模型,并比较两种不同方法的结果。方法将肾癌完整组织块和肿瘤细胞悬液种植于裸小鼠肾包膜形成原位移植瘤,并观察和比较两种方法所建立模型的肿瘤生长状况、移植成功率和自发转移的发生率。结果肾癌完整组织块接种的原位成瘤率达75·0%,肾、肺门淋巴结转移率均73·3%,肺转移发生率53·3%;细胞悬液种植的原位成瘤时间较组织块法延迟10d,成瘤率为40·0%,肺、肾门淋巴结转移率均12·5%,肺脏无转移。结论两种原位移植转移模型均具有人肾细胞癌自然生长过程的特点,肾癌完整组织块原位移植模型优于细胞悬液原位接种模型,为研究人肾细胞癌转移机制和抗转移实验治疗提供了有力工具。Objective To establish metastatic model of human renal clear cell carcinoma using orthotopic implantation of histologically intact tissue and cellular orthotopic injection of cell suspensions and compare their different resuits. Methods Using two methods to construct orthotopic nude mouse models: after RCC-949 passaged for 5 times by inoculating into nude mice sc, the tissue from the same xenotransplant tumor was used to construct surgical orthotopic implantation model. At the same time, the RCC-949 tumor cell suspension was injected orthotopically. The takeing rate, size of primary tumors and metastatic rate of human renal cell carcinoma RCC-949 were compared in two orthotopic nude mouse models. Results Histologically intact tissue model compared with the cellular orthotopic injection models: the take rate, pulomonary hilar lymph nodes metastatic rate, renal hilar lymph node metastatic rate and the lung metastatic rate were 75.0% vs 40.0% , 73.3% vs 12.5% , 73.3% vs 12. 5% 53.3% vs 0% respectively, while the size of primary tumors in the former model also larger and much more locally invasive. Conclusion The take rate and the metastatic rate in histologically intact tissue model are higher compared to the cellular orthotopic injection models. Hence histologically intact tissue model is appropriate for isolating metastic cells in vivo. This model should be of value in cancer research and antimetastatic drug discovery for renal cancer, a currently very poorly responding malignancy.
