共刺激分子B_7和CD_(28)在胃癌患者中的表达

摘　　要：目的:研究胃癌患者外周血中CD28表达阳性的T细胞数量和患者胃癌组织细胞表面B7的表达水平,以探讨胃癌患者共刺激通路是否异常。方法:采用流式细胞术检测胃癌患者外周血中CD28阳性T细胞与胃癌患者组织细胞B7分子的表达,并与胃良性疾病患者和正常人作比较。结果胃癌患者CD28阳性的T细胞数为(38.18±5.83)%,明显低于对照组(52.99±4.75)%,t=11.05,P=0.03。胃癌患者组织细胞表面B7表达水平为(50.60±7.48)%,低于对照组(64.51±5.93)%,P=0.04。结论:胃癌患者外周血中T细胞低表达CD28分子,胃癌细胞低表达B7分子,从而影响B7CD28共刺激通路,使T细胞不能有效地清除肿瘤。OBJECTIVE： To investigate costimulatory pathway of stomach cancer patients by examining peripheral CD28 posirive T lymphocytes and B7 molecules expression on the primary stomach cancer cells in patients with stomach cancer. METHODS： Flow cytometry was used to analysis CD28 and B7 molecules expressed on T lymphocytes and primary stomach cancer cells respectively. RESULTS： CD28 positive T lymphocytes in stomach cancer patients[（38. 18± 5.83）%] were lower than those in the control group [（52. 99±4. 75）%], t=11.05, P=0.03. B7 molecules ex pressed on primary stomach cancer cells [（50.60±7.48）%] were lower than those in control group [（64.51 ±5.93） %],P=0.04. CONCLUSIONS： Peripheral T lymphocytes in stomach cancer pa tients express low level of CD28 molecules. Primary stomach cancer cells express low level of B7 molecules. Costimulatory pathway dysfunction in stomach cancer patients results in T lymphocytes failure against the tumor cells.
