摘  要:目的:通过转染野生型p53基因致肝癌细胞株BEL-7404,研究其对肝癌细胞的生长及凋亡作用。方法:野生型p53基因通过脂质体Lipofectamine介导转染人肝癌细胞株BEL-7404。用四甲基偶氮唑(MTT)法测定细胞生长曲线,用流式细胞仪进行细胞凋亡分析。结果:肝癌细胞BEL-7404转染野生型p53基因后,其生长作用明显受到抑制,细胞生长抑制率在第4天可达50.8%。流式细胞仪分析细胞凋亡结果:阴性对照组为1.7%,转染pUHD10-3组为3.2%,转染pUHD10-3-p53组为40.7%。结论:野生型p53基因可诱导肝癌细胞BEL-7404的生长抑制及发生凋亡。Objective. To investigate the effect of p53 gene transfection on the growth and apoptosis ofhepatocarcinoma cell line BEL-7404. Methods: The HCC line BEL-7404 was transfected with plasmid pUHD10-3 recombined with wt-p53 gene mediated by Lipofectamine. 3-(4, 5-Dimethy-2-thiazolyl )-2, 5-diphenyl-2H-tetrazolium-bromid (MTT) technique was used to analyse the growth of HCC cells. Theapoptosis of the cells was evaluated by flow cytometry. Result:The growth of HCC cell line BEL-7404 wasinhibited after transfected with wt-p53 gene, and 50.8% of the cells were inhibited on the fourth day. The rates of apoptosis measured by flow cytometry were: 1. 7% in the untreated group, 3. 2%o in the group transfected with pUHD10-3, 40. 7% in the group tranfected with pUHD10-3-p53. Conclusion: The inhibition efficiency and apoptosis of hepatocarcinoma cell line BEL-7404 was induced by wt-p53 gene.
