摘 要:目的:探讨小鼠哮喘模型中T淋巴细胞对B淋巴细胞产生特异性IgG(sIgG)的调节作用。方法:雌性Balb/c小鼠40只,随机分成实验组(哮喘组)和对照组,每组各20只,实验组给予腹腔注射鸡卵白蛋白(OVA)6.7 mg.kg-1(第2~4周)及雾化吸入1%OVA,20 min.d-1(第5~6周),对照组注射同等剂量盐水及吸入无菌PBS。断头法处死小鼠,分离小鼠脾脏T、B淋巴细胞,分组进行细胞培养,以ELISA法检测各组细胞培养上清中sIgG的含量。结果:无论特异性抗原(OVA)是否参与,哮喘组小鼠B淋巴细胞sIgG分泌水平均明显高于对照组(P<0.05,P<0.01);哮喘组小鼠T淋巴细胞能使其自身B淋巴细胞sIgG分泌水平明显下降(P<0.05);抗原OVA刺激不能改变对照组和哮喘组小鼠B淋巴细胞自然状态下以及受刺激后的sIgG分泌水平(P>0.05);哮喘组小鼠T淋巴细胞能使对照组B淋巴细胞sIgG分泌水平升高(P<0.05);经哮喘组小鼠T淋巴细胞作用后,哮喘组鼠B淋巴细胞分泌sIgG水平低于对照组(P<0.05);对照组小鼠T淋巴细胞能降低哮喘组小鼠B淋巴细胞sIgG分泌水平(P<0.05)。结论:在支气管哮喘中,B淋巴细胞sIgG参与了哮喘发病,T淋巴细胞对B淋巴细胞分泌sIgG具有显著调节作用,T淋巴细胞在支气管哮喘发病中起重要作用。Objective To study the regulation of T lymphocytes on antigen-specific IgG (sIgG) from B lymphocytes in asthmatic mouse models and clarify the role of T lymphocytes in the pathogenesis of asthma. Methods Firstly asthma mouse models were established. Then the mouse-ventilation-function-testing system was used to select mice which were successfully immunized. T and B lymphocytes were isolated from the mouse spleen aseptically and were cultivated in different combination. Then the supernate was collected and the sIgG in the supernate were tested with enzyme-linked-immunosorbent-assay (ELISA). Results No matter if the specific antigen was involved, slgG secreted from B cells from asthmatic mice was higher than that of the B cells from control mice (P^0. 05 and P〈 0.01). After regulated by its own T lymphocytes, levels of slgG secreted by B cells from asthmatic mice decreased significantly (P〈0. 05). No matter if the T ceils from asthmatic mice was involved, the specific antigen couldn't change the levels of slgG secreted by B cells from asthmatic and control mice (P〉0. 05) ; after interfered by T lymphocytes from asthmatic mice, levels of slgG produced by B cells from normal control were increased obviously (P〈0. 05. ); after regulated with T lyrnphocytes from asthmatic mice, levels of slgG produced by B cells from asthmatic mice was lower than that of the B cells from control mice (P〈0. 05). With or without specific antigen, after regulated with T cells from asthmatic mice, levels of slgG produced from B cells of asthmatic mice decreased obviously (P〈0. 05 and P〈0. 01). Conclusion slgG is involved in the mechanism of asthma and T lymphocytes have a key role in regulating the pathogenesis of asthma. And T lymphocyte functional abnormality is one of the important mechanisms of asthma.
