摘  要:目的:通过观察颞叶癫痫大鼠海马CA1区超微结构变化,探讨颞叶癫痫的发病机制。方法:以海人酸杏仁核微量注射建立经典的雄性Wistar大鼠颞叶癫痫化学点燃模型,按点燃后第14、21天时间点分为2组,每组5只,观察大鼠海马CA1区神经细胞超微结构,并对突触活性参数量化分析。结果:颞叶癫痫大鼠海马CA1区神经细胞结构受损,神经元突触数密度降低(P<0.05),突触活性区膜面积缩小(P<0.05),突触界面曲率降低(P<0.05),比表面减小(P<0.05),突触小泡数密度降低(P<0.05)。星形胶质细胞线粒体体积密度增加(P<0.05),数密度增高(P<0.05)。结论:颞叶癫痫大鼠海马CA1区神经细胞和突触超微结构存在远期损害和活力降低。Objective To investigate the mechanism of temporal lobe epilepsy through observing ultrastructural changes of CA1 region in hippocampus in temporal lobe epileptic male rats. Methods Temporal lobe epileptic typical models were established in Wistar male rats kindled by kainic acid microlnjected into right Nucleus amygdalae. Ultrastructural observation; on nerve cells and synapses in CA1 region were performed, as well as quantitative analysis on their active parameters on 14 d, 21 d after kainic acid injection in five rats, respectively. Results There existed nerve cells structural damage in CA1 region in temporal lobe epileptic rats, synaptic parameters including synaptic density, synaptic active area, synaptic interface curvature, surface ratio, synaptic vesicles in epileptic group were significantly decreased than those in sham group (P〈0. 05). The number or volum of mitochondria in astrocytes in epileptic group were significantly increased than those in sham group (P〈0. 05). There existed long-term ultrastructural and active paramete changes in nerve cells and synapses. Conclusion There are structural and functional injuries in nerve cells and synapses in CA1 region in temporal lobe epileptic male rats.
