摘 要:目的评价张应力作用后面颌肌细胞烟碱样乙酰胆碱受体基因表达变化。方法采用已经建立的面颌部肌细胞体外力学刺激—细胞培养模型,半定量RT-PCR技术检测nAChRα1亚基基因加力前后转录水平变化。结果未发现有明显统计学差异。结论单纯张应力对体外培养小鼠颌面部骨骼肌细胞nAChR基因表达影响较小。Objective:To elucidate the effect of strain on nicotinic acetylcholine receptor (nAChR) mRNA expression in maxillofacial skeletal myocytes. Methods : Skeletal myocytes of BALB/C mice were cultured and strained by a straining device, Focel, with the force of 0.1 % cell deformity for 2,4,6,8 and 12 h respectively, Then the cells were collected,nAChRα1 subunit mRNA expression was examined by Semi-quantitative RT-PCR test. Results:No significant difference was found in nAChtRα1 subunit mRNA expression between strained and unstrained cells( P =0, 30) , no difference was found among the cells treated for different times( P = 0.13 ). Conclusion:Strain at the force of 0. 1% cell deformity has no effect on the nicotinic acetylcholine receptor (nAChR) expression in maxillofacial skeletal myoeytes of mouse in in vitro.
