摘 要:目的:观察诱导型和神经型一氧化氮合酶抑制剂对脊髓损伤大鼠神经传导功能恢复的影响。方法:实验于2003-04/05在南方医科大学动物实验中心完成。选取清洁级雄性SD大鼠40只,随机分为损伤对照组、氨基胍组、7-硝基吲唑组和正常对照组,每组10只。制备脊髓压迫伤模型,氨基胍和7-硝基吲唑剂量均为100mg(/kg·次),氨基胍给药时间为伤前1h、伤后8,24,36h4次,7-硝基吲唑给药时间为伤前30min和伤后4h共2次,均为腹腔注射。给药24h后每组随机选取4只动物采用双缩脲法测定检测一氧化氮含量和一氧化氮合酶活性。其余动物4周后用电生理学(体感诱发电位和运动诱发电位检测)和核磁共振和动物行为学(动物行为学进行BBB评分观察最高21分,最低0分)等指标评价神经功能的恢复情况。结果:各组实验动物完成全部实验,无脱落。均进入结果分析。①正常组织中即存在一氧化氮含量和一氧化氮合酶,损伤后呈升高趋势,以损伤对照组最明显,与其他3组相比差异明显(P<0.05)。7-硝基吲唑组和氨基胍组可使一氧化氮合酶活性下降,前者作用明显高于后者(P<0.05);7-硝基吲唑组和氨基胍组可使一氧化氮含量下降,氨基胍组强于7-硝基吲唑组。②7-硝基吲唑组和氨基胍组动作电位较损伤对照组有明显恢复,而损伤对照组则基本没有明确的波形出现;7-硝基吲唑组和氨基胍组及正常对照组潜伏期和振幅比损伤对照组有明显差异(P<0.05);7-硝基吲唑组和氨基胍组两组之间比较则差异不明显。损伤后体感诱发电位基本没有明显恢复。③伤后MRI信号(T2)均存在不同程度的改变,如脊髓的前后径变窄,信号强度增高等,伤段脊髓组织存在广泛的变性和纤维化,正常信号连续性中断,尤以损伤对照组组最明显,而治疗组则表现有比较连续的信号。④氨基胍组和7-硝基吲唑组与损伤对照组相比,BBB评分结果明显提�AIM: To observe the effects of induced neural nitricoxide synthase (NOS) inhibitor on the recovery of nerve conduction function in rats with spinal injury. METHODS: The experiment was done in the Animal Experiment Center, Southern Medical University from April to May 2003. Forty clean grade male SD rats were selected and divided randomly into injured control group, aminognanidin group, 7-nitroindazole (7-NI) group and normal control group with 10 in each group. Spinal cord compression injury model was made, and the dosages of aminoguanidin and 7-NI were both 100 mg/kg per time. The administration time of aminoguanidin was at 1 hour before injury, 8, 24 and 36 hours after injury. The administration time of 7-NI was at 30 minutes before injury and 4 hours after injury. Both were intraperitoneal injection. At 24 hours after administration 4 animals were selected randomly in every group to detect the content of nitrogen monoxide and activity of NOS with biuet. The recovery condition of the neural function of the other animals was detected by eilectrophysiology (somatosensory evoked potential and movement induced potential detection), magnetic resonance imaging and animal ethology (animal ethology performing BBB score observation: the highest 21 points, the lowest 0 point) etc. index after 4 weeks. RESULTS: The animals in every group were experienced the whole experiment without drop, and all involved in the result analysis. ① In the normal tissue there was the content of nitrogen monoxide and NOS, which showed the trend of increasing after injury. That in the injured control group was the most significant, and the difference was significant compared with other 3 groups (P 〈 0.05). The activity of NOS decreased in the aminoguanidin group and 7-NI group, and the effect in the former was higher than that in the latter (P 〈 0.05); The content of nitrogen monoxide decreased in the 7-Nl group and aminoguanidin group, and it was better in the aminoguanidin group than that in the 7-NI gr
