摘 要:择到一株产尿酸酶较高水平的产朊圆酵母(Torula utilisHenneb).对此菌株尿酸酶形成条件的研究表明:蔗糖、葡萄糖是酶形成的适合碳源;蛋白胨和酵母膏是产酶的适合氮源,另添加无机氮源对产酶略有帮助作用;尿酸、黄嘌呤和鸟嘌呤对酶形成起诱导作用,底物尿酸是适宜的诱导物.尿酸酶形成最适培养基组成为(%):蔗糖3,蛋白胨2,酵母膏0.5,(NH4)2SO40.05,尿酸0.05,KCl 0.04,Pb2+、Fe3+、Hg2+、Mg2+、Ag+、Ca2+、Zn2+不利于产酶.另对培养条件做了优化探索:最适pH为6.0,在250mL三角瓶中装30mL培养液,接种量10%,在200r/min的旋转摇床上28℃振荡培养23h.A Torula utilis Henneb with high performance of producing uricase was screened out and the uricase-forming conditions was investigated. The results were obtained as follow: Sucrose and glucose were the suitable carbon sources and proteose-peptone and yeast extract were the suitabe nitrogen sources for enzyme formation. In addition, addition of inorganic nitrogen sources enhanced enzyme production. Uric acid, xanthine and guanine were found to be effective inducers for enzyme formation. Uric acid, the substrate of the uricase, was the best inducer. The best medium for uricase formation was consisted of sucrose 5 %, proteosepeptone 2%, yeast extract 0.5%, (NH4)2SO4 0.05%, uric acid 0.05%, KCl 0.04%. Pb^2+、Fe^3+ ,Hg^2+ , Mg^2+ , Ag^+ ,Ca^2+ and Zn^2+ went against enzyme formation. Furthermore, the author did some research about the condition of fermentation. The optimum temperature was 5.5. When the yeast was grown in 250 mL conic flask containing 30 mL of the medium mentioned above on the rotary shaker (200r/min) at 28℃ for 23h,uricase with maximal activity was obtained.
