摘  要:目的观察体外培养的HT-29细胞在丁酸钠作用后形态学及生长曲线的变化。初步探讨丁酸钠对结肠癌细胞的凋亡诱导作用。方法利用相差显微镜、可变投射显微镜、电镜观察细胞形态学变化,MTT法检测HT-29细胞生长情况。结果丁酸钠作用后的HT-29细胞形态发生改变,表现为细胞体积变小、形状变圆,细胞核固缩,脱壁细胞逐渐增多,细胞膜突起减少,核内染色质呈团块状积聚于核膜下,并可见凋亡小体。丁酸钠2.5、5和10mM对HT-29细胞生长均有抑制作用,并呈浓度、时间依赖性。结论细胞形态学的变化初步显示了丁酸钠对结肠癌细胞具有凋亡诱导作用。Objective Observing the change of configurition and growth curve of HT -29 cells treated with sodi- um butyrate and the effect of sodium butyrate on induction of apoptosis in colonic neoplasm cells. Methods Cells were observed by PCM, BVPM, EM.MTT examine the growth of HT - 29 cells. Results Configurition of HT - 29 cells were changed after treatment with sodium butyrate. The cells became smaller, shape of cells rounder and pyknosis, shedding cells increased gradually. The number of protrusion of cell membrane decreased. Chromatin accumulate under the nuclear envelope . Apoptosis bodies appeared also by EM. 2.5,5 and 10mM sodium butyrate all can cause growth - inhibition. The effects were time - and dose - dependent. Conclusion Sodium butyrate have an effect on induction of apoptosis in colonic neoplasm cells.
