食管癌p63基因mRNA的异常表达

摘　　要：目的研究p63基因mRNA在原发性食管鳞癌的表达情况,了解该基因在原发性食管鳞癌发生、发展和预后中的意义。方法采用RT-PCR检测了38例原发性食管鳞状上皮癌及对应的正常组织p63基因mRNA的表达情况。结果73.68%(28/38)原发性食管鳞状上皮癌检测到ΔNp63基因mRNA的表达,而对照组38例正常组织中有30例(78.94%)表达ΔNp63基因mRNA,其中低分化癌的表达率高于高、中分化癌(P<0.05),而与年龄、性别、淋巴结转移和临床分期无关。全部样本中仅1例检测有TAp63mRNA的表达。结论p63在食管癌和上皮细胞中表达的亚型主要是ΔNp63,其在低分化组的高表达,说明ΔNp63参与了食管鳞癌的发生、发展,并与食管鳞癌的预后有关。Objective To explore the penetrance of p63 in esophageal squamous cell cancer. Methods p63mRNA expression in squamous cell carcinomas and histologically normal mucosa of the esophagus were analyzed by reverse transcriptase - polymerase chain reaction （ RT - PCR）. Results The DeltaNp63 mRNA was detected in all malignant and histologically normal tissues, positive rates were respectively 73.68% （ 28/38 ） and78.94% （ 30/38 ）. Whereas TAp63 presented extremely low or no expression and only 1 sample was positive in all samples. Expression of the DeltaNp63 mRNA was higher in low - differentiated group than well and moderate - differentiated group, without significant difference in age, gender, lymph node metastasis and stage. Conclusion DeltaNp63 is the major isotype expressed in epithelia cancers of the esophagus. Elevated expression of DeltaNp63 mRNA may play a significant role in the development and prognosis of the esophageal squamous cell carcinoma.
