摘  要:目的:研究慢性神经痛大鼠脊髓缝隙连接蛋白43(Connexin43,Cx43)基因表达的变化.方法:18只成年SD大鼠随机分为两组(n=9),分别接受右侧坐骨神经结扎手术(CCI组)和假手术(Sham组).手术前、后观察大鼠疼痛行为学并测定右后肢热刺激抬脚潜伏期(PWTL)、电刺激抬脚阈值(PWET)的变化,于术后14天取大鼠L4/5右侧脊髓,以β-肌动蛋白(β-actin)为内参照基因,采用RT-PCR半定量分析CCI组和Sham组Cx43表达的差异.结果:CCI组大鼠表现为行走步态异常,与Sham组相比CCI组术后5~14天疼痛阈值明显降低(P<0.01).Sham组Cx43扩增产物量极少,而CCI组L4/5 Cx43扩增产物量约为Sham组的3倍.结论:慢性神经痛大鼠脊髓Cx43基因表达显著增加,提示脊髓缝隙连接可能在慢性病理性痛过程中发挥重要的作用.Objective: To investigate the alteration of Cx43 gene expression in rat spinal cord with chronic neuropathic pain. Methods: 18 adult SD rats were randomly divided into 2 groups (n = 9), and received sciatic nerve chronic constriction injury (CCI group) and sham-operation (Sham group) respectively. The paw withdrawal thermal latency (PWTL) and paw withdrawal electric threshold (PWET) were assessed. Fourteen days after operation, the right spinal L4/5 segments were removed, and the difference of Cx43 gene expression between two groups was analyzed with semi-quantitative RT-PCR, according to the expression of β-actin. Results: In CCI group, most of the rats showed pain-related behaviors. The PWTL and PWET in CCI group were significantly lower than that in Sham group(P 〈 0.01). The RT-PCR amplified product for Cx43 was rarely seen in Sham group, and those in CCI group was 3 times as much as that in Sham group. Conclusion: There was a remarkable increase of Cx43 gene expression in the spinal cord of rats with chronic neuropathic pain, indicating that gap junction may play an important role in the pathophysiologic process of chronic neuropathic pain.
