摘 要:目的:研究硒蛋氨酸对肝癌细胞株SMMC7402生长的影响及其作用机制。方法:采用MTT比色法、流式细胞术、DNA片段分析法,研究硒蛋氨酸对肝癌SMMC7402细胞生长、增殖及凋亡的影响。结果:硒蛋氨酸呈时间-剂量依赖性方式抑制SMMC7402细胞生长增殖;G1期前出现亚倍体凋亡峰,细胞阻滞于G0/G1期;DNA电泳出现特征性凋亡梯带。结论:硒蛋氨酸可抑制肝癌细胞株SMMC7402的生长增殖,诱导细胞凋亡。Objective To evaluate the effect of selenomethionine (SeMet) on the apoptosis of hepatocellular Carcinoma cell line SMMC7402 and explore its mechanism. Methods Hepatocellular Carcinoma cell proliferation and apoptosis were determined by MrIT assay and cell growth curve, flow cytometry and DNA agarose gel electrophoresis ,respectively. Results SeMet caused gzowth inhibition and apoptosis time- dependently and dose- dependently in SMMC7402 cells, and caused an increase in arrest in the G0/G1 phase of the cell cycle, Flow cytometry analysis revealed an apoptotic peak ahead of G1 phase. DNA agarose gel electrophoresis showed DNA fragmentation. Conclusion SeMet may be a new therapeutic pathway for hepatocellular carcinoma.
