摘  要:目的:评估大蒜素与B7·1转基因肿瘤疫苗对膀胱癌的协同抗癌效果。方法:脂质体法转染鼠膀胱肿瘤(MBT-2)细胞。流式细胞术测B7·1基因的转染效果。相差显微镜观察大蒜素对肿瘤细胞的细胞毒作用。动物实验检测联合治疗的抗癌效果。组织切片检测肿瘤组织中免疫反应的发生。LDH释放法测治疗后淋巴细胞对肿瘤细胞的细胞毒活性。结果:约40%的转染细胞表面能表达B7·1分子。大蒜素对膀胱肿瘤细胞有直接的细胞毒作用。当B7·1转基因肿瘤疫苗与大蒜素联合应用时,具有最大的抗肿瘤效果。结论:大蒜素与转基因肿瘤疫苗具有良好的协同抗肿瘤作用。Objective To evaluate the anti-tumor effect of allicin combined with B 7·1 modified tumor vaccine Methods Murine bladder cancer cell line, MBT-2, was transduced with lipofectine and B 7·1 plasmid. The transfection efficiency was confirmed by low cytometry. The cytotoxicity of allicin was measured by observing the morphology of tumor cells. Anti-tumor activity of combination gene therapy was measured by animal experiment. Tumor tissues were examined by hematoxylin and eosin staining. CTL activity of lymphocyte was measured by the release of endogenous lactate dehydrogenase(LDH). Reauits Allicin showed an obvious cytotoxicity to bladder tumor cells. Combination Allicin- B 7·1 gene therapy showed the most significant anti-tumor activity compared with the other groups. There were more macrophages and lymphocytes at the tumor margin and extensive necrosis in the center of the tumor that was treated by allicin combined with B 7·1 modified tumor vaccine than the tumor in control group. Vaccination with allicin and MBT-2 B 7·1 generated the strongest cytotoxic lymphocyte responses to parental MBT-2 tumor cells. Conclusion Allicin treatment combined with gene modified tumor vaccine had significant additive anti-tumor effects.
