通络益智方对血管性痴呆大鼠脑内超氧化物歧化酶和一氧化氮的影响

摘　　要：目的观察自拟通络益智方对血管性痴呆大鼠脑内超氧化物歧化酶(SOD)活性和一氧化氮(NO)含量的影响,探讨该方治疗血管性痴呆的作用机理。方法经颈外动脉注入血凝栓子加山莨菪碱皮下注射,造成血管性痴呆大鼠模型,以记忆力试验检测模型,另测量各组大鼠脑内皮层、丘脑、海马、纹状体部的SOD活性和脑内及血清NO含量。结果给药组与模型组比较,大鼠学习记忆力明显增强,脑4部位SOD活性和NO含量增加(P<0.01或P<0.0.5),血清NO无明显改变。通络益智方对脑皮层SOD、NO含量的影响明显强于对照药三乐喜(P<0.01),该方对脑其余3部位的SOD、NO的影响与三乐喜相当(P>0.05)。结论通络益智方具有提高血管性痴呆动物脑内SOD活性,增加NO含量,改善脑功能的作用。Objective To observe the influence of Tongluoyizhi Formula （TLYZ） for activation of superoxide dismutase （SOD） and nitric oxide （NO） in brain in order to explore the mechanism of TLYZ treating vascular dementia. Methods Injecting embolism to external carotid artery and giving Anisodaminte by hypodermic to make models of rat vascular dementia, then detect the model by anamnesis experiment and measured the activity of SOD and the contents of NO in pallium the thalamic, hippocampus and striatum of rats and the contents of NO in serum. Results To compared with control group, the ability of learning and memory was significantly enhanced in treating group, the activity of SOD and contents of NO increased significantly （P 〈 0.01 or P 〈 0.05）, but the contents of NO in serum has no significant change. The influence of TLYZ for activation of SOD and NO in pallium was significantly different from the Aniracetam. TLYZ was similar to Aniracetam on influence for activation of SOD and NO on other three parts of cerebrum （P 〉 0.05）. Conclusion TLYZ has the function of increasing the activation of SOD and the contents of NO. It has also ameliorated the function of brain.
