大鼠局灶性脑缺血再灌注后TACE mRNA表达变化的研究

摘  要:目的研究大鼠局灶性脑缺血再灌注后不同时间脑皮质半暗带和中心区肿瘤坏死因子转换酶(TACE)转录水平的表达规律.方法用线栓法建立大鼠大脑中动脉缺血再灌注模型,切取缺血半暗带及中心区皮质组织,采用逆转录-聚合酶链反应(RT-PCR)测定TACE mRNA水平的变化.结果在脑皮层缺血半暗带及中心区脑组织,TACE mRNA表达变化有两个高峰,分别在脑缺血再灌注后3 h、12~24 h(P<0.01),在脑缺血再灌注后72 h表达低于正常对照组(P<0.05或P<0.01),7、14 d未检测到TACE mRNA水平,21 d出现低水平表达.结论TACE mRNA过度表达可能对脑缺血再灌注后早期神经元死亡起促进作用.Objective To investigate the expression of tumor necrosis factor-α converting enzyme transcription level in rat's focal cerebral ischemic penumbra and core of cortices at different ischemic time. Methods The middie cerebral artery occlusion models were established by using the intraluminal suture occlusion method in Wistar rats. Brain samples were harvested from ischemic penumbra and core of cortices. The change of TACE mRNA was assessed by RT-PCR. Results Expression of TACE mRNA in the penumbra and core regions reached the peak at reperfusion 3 and 12-24 h ( P 〈 0.01 ), rapidly decreased to the low level in 72 h following ischemic reperfusion ( P 〈 0.05 or P 〈 0.01) ,and wasn't detected in 7 d and 14 d. It came back to the low level in 21 d. Conclusion The overexpression of TACE mRNA was notably induce neuronal death after focal cerebral ischemic-reperfusion.
