HE切片中苏木素沉渣去除方法探讨

摘　　要：目的:探讨去除HE染色切片中苏木素沉渣的方法。方法:采用下列三种方法去除HE染色切片中的苏木素沉渣并比较其效果:(1)陈旧苏木素染色液加热至40℃;(2)滤纸过滤陈旧苏木素染色液;(3)石蜡切片经1.0%NaOH浸泡8min、1.0%NaHCO3浸泡2min、200U/ml糜蛋白酶浸泡8min后在陈旧苏木素染色液中染色。此外,在AB/PAS染色前和染色后用1.0%NaOH、1.0%NaHCO3、200U/ml糜蛋白酶浸泡切片,观察该化学处理对粘液染色效果的影响。结果:经过1.0%NaOH、1.0%NaHCO3或200U/ml糜蛋白酶浸泡的切片,HE染色后无苏木素沉渣附着;陈旧苏木素染色液加热后染色,切片中仍有大量苏木素沉渣附着,过滤后染色则切片中苏木素沉渣消失。在AB/PAS染色后、苏木素衬染前1.0%NaOH、1.0%NaHCO3或200U/ml糜蛋白酶处理切片,粘液区呈强阳性染色;AB/PAS染色前用上述方法处理切片,粘液染色强度略有减弱。结论:陈旧苏木素染色液过滤经1.0%NaOH、1.0%NaHCO3、200U/ml糜蛋白酶浸泡均可有效去除HE染色切片中的苏木素沉渣。Objective： To explore the good methods of elimination of hematoxylin sediments in HE sections. Methods： Three methods as follow were employed to eliminate the hematoxylin sediments in HE sections： 1. Over - used hematoxylin solution was heated to 40℃ ; 2. Over - used hematoxylin solution was filtrated by filter paper; 3, The sections were marinated in 1% NaOH for 8 min, in 200 U/ml chymotrypsin or 1.0% NaCO3 for 8 min for 2 min before the step of over - used hematoxylin staining. Pre - marinated the sections in 1. 0% NaOH, 200 U/ml chymotrypsin or 1.0% NaHCO3 were used for AB/PAS stain, and the staining intensity was compared with the sections which has marinated in 1.0% NaOH, 200 U/ml chymotrypsin or 1.0% NaHCO3 after AB/PAS stain. Results： No hematoxylin sediment was seen in the HE sections which were stained in filtrated over - used hematoxylin solution, but a lot of hematoxylin sediments were present in the HE sections which were stained in heated over - used hematoxylin solution. There was also no hematoxylin sediment in the HE sections which were pre - processed in 1. 0% NaOH, 200 U/ml chymotrypsin or 1. 0% NaHCO3. Strongly positive AB staining was showed in the mucus area of the sections which were marinated in 1.0% NaOH, 200 U/ml chymotrypsin or 1.0% NaHCO3 after the step of AB/PAS staining. However, the intensity of AB staining had a little bit reduction in those sections which was pre-marinated in 1.0% NaOH, 200 U/ml chymotrypsin or 1.0% NaHCO3 before the step of over - used hematoxylin staining. Conehlsion： Filtrating over - used hematoxylin solution or pre - marinating sections in 1.0% NaOH, 200 U/ml chymotrypsin or 1.0% NaHCO3 can effectively eliminate hematoxylin sediments in HE sections.
