胃炎消对胃癌癌前病变患者细胞周期素依赖性激酶及PCNA和Ki-67蛋白表达的影响

摘　　要：[目的]探讨胃炎消逆转胃癌癌前病变(PLGC)的作用机制。[方法]采用链霉素抗生物素蛋白-过氧化酶(LSAB)免疫组化法检测胃炎消治疗组和维酶素对照组共35例PLGC患者治疗前后细胞周期素依赖性激酶(cy-clin-dependent kinases,CDK4)、相关周期蛋白PCNA抗原及Ki-67抗原的变化。[结果]CDK4蛋白在各胃黏膜的阳性表达率普遍较高,经胃炎消治疗后,CDK4蛋白的表达率及表达强度变化不大,治疗前后比较差异无统计学意义(P>0.05);PCNA抗原和Ki-67抗原在胃黏膜病变中的蛋白表达均随着上皮细胞增生程度的增加和肠上皮化生的出现而递增,两种抗原并呈现显著的正相关性,胃炎消组治疗后PLGC细胞增殖指数较治疗前明显降低(P<0.05),而维酶素对照组则无明显变化(P>0.05)。[结论]胃炎消有抑制PLGC组织细胞过度增殖的作用,并可能是通过纠正PLGC细胞增殖而起到治疗作用。[Objective] To explore the acting mechanism of Weiyanxiao （WYX） in the treatment of precancerous lesions of gastric cancer （PLGC）. [Methods] Thirty-five cases of PLGC were divided into two groups： WYX-treated group and Weimeisu-treated group. The protein expression of CDK4, PCNA, Ki-67 in all cases was detected by immunohistochemistry respectively. [Results] The expression of CDK4 protein was observed in certain cases of PI.GC and increased with the progress of lesions. Neither WYX nor weimeisu had effect on it. The overexpression of PCNA and Ki-67 proteins was detected in certain eases of PI.GC and increased with the progress of lesions. WYX significiently inhibited the expression of PCNA and Ki-67 proteins. [Conclusion] WYX can promote CDK4 to combine with p16 and CyclinD1 competitively, so as to weaken pRb phosphorylation acid and reverse the over-proliferation of PI.GC through regulating the expression of P^16 and CyclinD1 protein.
