摘  要:目的研究BRCA1、MMP-2、PTEN、CD44v6、nm23-H1、Cath-D在乳腺癌组织中的表达及生物学意义,筛选与预后有关的最佳因子组合。方法采用免疫组化S-P法标记上述指标。结果(1)乳腺癌组织中PTEN、BRCA1、nm23-H1蛋白表达明显降低,表达水平与复发、淋巴结转移呈负相关,与生存期呈正相关。(2)CD44v6、MMP-2、肿瘤间质内Cath-D的蛋白表达增高并与淋巴结转移、复发呈正相关。(3)BRCA1蛋白与nm23-H1、PTEN蛋白表达呈正相关,与MMP-2表达呈负相关。MMP-2蛋白表达与CD44v6表达呈正相关。结论乳腺癌组织中MMP-2表达增强、nm23-H1和BRCA1表达抑制,可能作为预测肿瘤转移及判断预后的可靠指标。Purpose To investigate the expression of nm23-H1, BRCA1, PTEN, eathepsin D, MMP-2,CD44v6 proteins and their clinical significance in human breast cancers. Methods The expressions of biological marker proteins were determined by immunohis-toehemieal S-P staining in 106 cases of infiltrating breast cancers. Results In breast cancers, the expressions of CD44v6, MMP-2, cathepsin D were increased ,the expression of nm23-H1, PTEN were decreased. The positive expressions of CD44v6, MMP-2 and the negative expression of nm23-H1, PTEN, BRCA1 were positively correlated with recurrence. There was a statistically significant correlation between eathepsin D expression of stromal cells and recurrence. In breast cancers, the expression of BRCA1 had positive correlation to that of nm23-H1, PTEN, and had negative correlation to that of MMP-2. The expression of MMP-2 had positive correlation to that of CD44v6. Conclusions Enhancement of MMP-2 expression, inhibition of nm23-H1 and BRCA1 expression may he the reliable markers for predicting tumor metastasis and prognosis.
