摘  要:提高不同长度基因片段的分辨率是基因分析的关键问题,我们对电泳分辨基因差异片段的方法在制胶、染色及凝胶保存技术上进行了三个方面的改进,建立了"凝胶干燥长期保存法",能客观反映实验结果,重复性好,对一些目前尚未认识的片段,留下了原始档案资料,供将来进一步分析;干燥胶片还可照像、投影、复印和激光扫描。Distinguishing the differential gene fragments is the key of gene analysis.Traditional methods are loading DNA on agarose gels electrophoresis,stainning with ethidium bromide and observation on ultraviolet light.Sometimes the electropherosis bands of small DNA fragments can not be distinguished very clearly.We developed some technique involved polyacrylamide gels preparing,gels staining as well as gels preserving,and made up a new method to dry gels for long time preserving.Using the new method,polyacrylamide gelscan be dried and kept true situation for long time after assay and conserved as original files.Some PCR unspecialized fragments which can not be recognized at present could be analysed in future.The dry gels can also be used for photograph,projection,copy and laser scanning directly.
