摘　　要：目的构建人源抗肝癌单链抗体。方法从肝癌患者外周血中分离淋巴细胞,利用噬菌体展示技术,构建人源抗肝癌噬菌体抗体库并筛选人源抗肝癌单链抗体,用ELISA方法检验该抗体与抗原结合活性。结果成功构建了库容为4.0×106的人源抗肝癌单链抗体库,初步筛选到与肝癌细胞特异性结合的单链抗体。结论成功构建人源抗肝癌单链抗体,为进一步临床应用奠定基础。Objective To construct human single-chain Fv antibody against hepatocellular carcinoma. Methods Lymph cell was from the blood of the patients with hepatocellular carcinoma. The scFv gene library was constructed by phage display technology. The affinity to antigen was detected by ELISA after panning. Results The human single-chain Fv antibody library of hepatocellular carcinoma was successfully constructed. ELISA revealed that the scFv could specifically conjugate with HCC antigen. Conclusion Human single-chain Fv antibody of hepatocellular carcinoma was successfully constructed.
