基质溶解因子基因在人直肠癌中的表达及其临床意义

摘　　要：目的通过检测基质溶解因子(MMP-7)基因在直肠癌中的表达,探讨其在直肠癌进展中的作用。方法86对直肠癌肿瘤组织与正常直肠黏膜组织标本自身配对分成肿瘤组与正常组,提取总RNA,采用实时荧光定量逆转录-多聚酶链反应(Quantitative-Real-Time RT-PCR)法检测MMP-7基因的表达。结果MMP-7基因表达比值大于1的52例(62.7%,52/83),在肿瘤组的表达水平高于正常组(P<0.01)。MMP-7基因表达比值大于1的例数在Dukes C+D期与Dukes A+B期中分别为34例(72.3%,34/47)和18例(50.0%,18/36,P<0.05)。MMP-7基因表达与组织学分化程度(r=-0.635)和CEA水平(r=0.580)相关。结论MMP-7基因在直肠癌中的表达上调。MMP-7在直肠癌的浸润与转移中具有重要作用。Objective To detect the MMP-7 gene expression in human rectal cancer and its correlation with invasion and metastasis of human rectal cancer. Methods Eighty-six paired samples of rectal cancer and distant normal rectal tissue were allocated into cancer group and normal group. Total RNA extraction and relative Quantitative-Real-Time RT-PCR were proceeded. Results The expression ratios of MMP-7 between two groups were over 1 in 52 samples （62.65%）. The expression level of MMP-7 in cancer group was higher than that in normal group （P〈0.01）. The expression ratios were over 1 at Dukes C＋D and Dukes A＋B stage in 34 （72.3% ,34/47） and 18 （50.0%,18/36, P〈0.05） respectively. The expression level of MMP-7 gene was correlated with histological differentiation stage （r = - 0.635） and CEA level （r = 0.58）. Conclusion The expression level of MMP-7 is up-regulated in rectal cancers. MMP-7 plays an important role in invasion and metastasis of human rectal cancer.
