摘  要:目的:探讨乙酰肝素酶(HPA)反义寡核苷酸(AS-ODN)对CD44v6表达的影响。方法:采用脂质体介导HPAAS-ODN转染人胃癌细胞株SGC-7901,RT-PCR检测HPAmRNA的表达,采用免疫细胞化学SABC法观察细胞内CD44v6表达的变化。结果:脂质体介导转染HPAAS-ODN48h后MSGC-7901胃癌细胞内HPAmRNA表达下降MCD44v6阳性表达率转染前69.4%M转染不同浓度的AS-ODN后分别为56.5%,54.1%,46.5%和41.3%。转染后细胞CD44v6表达明显减少(P<0.01)。结论:HPAAS-ODN可以有效抑制胃癌细胞CD44v6的表达。Objective: To evaluate the effect of heparanase antisense oligodeoxynucleotide(ASOND) on the expression of matrix metalloproteinase-2 (MMP-2)and tissue inhibitor of metalloproteinase-2 (TIMP-2)in human gastric carcinoma cell line SGC-7901. Methods: Human gastric adenocarcinoma cell line SGC-7901 was transfected with liposome-mediated heparanase AS-OND. The expression of heparanase mRNA was detected after transfection by semi-quantitative total RNAs RT-PCR. The changes of MMP-2 and TIMP-2 were observed by immunocytochemistry. Results: The expressions of heparanase mRNA and MMP-2 of SGC-7901 cells treated with AS-ODN of different final concentrations were significantly decreased 48 hours later compared with those of the controls (P〈0.01). Conclusion: Heparanase AS-ODN has a significant inhibitory effect on the expression of MMP-2 in human gastric carcinoma cell line SGC-7901 .
