摘 要:探讨了胸腺素α1在小鼠S180肉瘤中的表达及其临床意义,以小鼠肉瘤S180组织和正常小鼠组织为材料,福马林固定和石蜡包埋,运用兔抗胸腺素α1多克隆抗体,按常规免疫组织化学方法检测接种在小鼠皮下的小鼠肉瘤组织以及正常小鼠皮下肌肉组织细胞中胸腺素α1的表达状况.研究结果发现胸腺素α1定位表达在小鼠肉瘤组织细胞的胞浆和细胞核中,免疫组化呈现强阳性,而正常组织细胞内基本无显色,呈阴性.试验结果表明:胸腺素α1在恶性肿瘤细胞小鼠肉瘤组织细胞中高表达,而正常小鼠组织则低表达或不表达.细胞内胸腺素α1蛋白的高表达与肿瘤具有明显相关性,该研究成果可能使胸腺素α1作为一个新的和有价值的肿瘤标记物在临床诊断中得到应用,也为进一步探索胸腺素α1与肿瘤之间相互关系的分子机理研究奠定基础,并通过抑制胸腺素α1这条途径达到控制肿瘤生长的效果.The objective of this study is to detect thymosin α1 expression in transplant 180 of mice using immunohistochemical (IHC) staining,and also to evaluate the relationship between Tα1 expression level and cancer. The S180 tissue and the normal tissue taken from mouse were treated with formalin fixed, paraffin-embedded, and analyzed by normal immunohistochemical analysis for detecting the expressed of Tα1, the rabbit antibody to Tα1 was acted as the primary antibody in immune analysis. The results of experiment showed that Tα1 was over expressed in S180 cancer cells and the staining pattern of cytoplasmic and nucleus showed strong positive,whereas the normal tissue cells of mice showed negative that was weak stained or did not stain at all. The experimental result proved that there was obviously positive correlated between the over expression of Tα1 of cell and that of the malignant cancer,which indicated that a positive expression of Tα1 was one of useful and new tumor marker for clinical diagnosis of malignant cancer,and the Tα1 may thus be acted as a significant factor for predicting a favorable prognosis. Based on this result,it will help to establish a foundation for further studying the mechanism about the highly expression of Tα1 and malignant cancer on the level of molecular. Then stressed pathway mediated by Tα1 may therefore be responsible for controlling the growth of cancer cells.
