健乳灵对乳腺增生大鼠乳腺组织p16、p53表达的干预作用

摘　　要：目的研究健乳灵对乳腺增生大鼠乳腺组织中抑癌基因p16、p53表达的干预作用。方法用苯甲酸雌二醇和黄体酮制作乳腺增生病大鼠模型,分别用不同剂量的健乳灵治疗60 d,用免疫组化法测定乳腺组织的p16、p53表达水平。结果健乳灵能抑制乳腺上皮增生,降低大鼠乳腺增生组织的p16、p53蛋白表达水平,与模型组比较有显著性差异(p<0.01)。结论健乳灵对大鼠的乳腺增生病有调节和治疗作用。Objective To Study the Effect of a Chinese traditional medicine “Jiannding” on the Expression of p16 and p53 in mammary gland hyperplasia in rats. Methods 75 female rats were randomly divided into 5 groups： normal control, hyperplasia model control, Jiaruling-treated group Ⅰ , Ⅱ and Ⅲ, 15 rats per each group. The rat model of mammary gland hyperplasia was established by injection with estradiol and progesterone. The level of expression of p16 and p53 were determined by immunohistochemical staining after treatment with Jianruling at a dose of 0.842g/kg/d, 1.684g/kg/d, 3.368g/kg/d for 60 days, respectively. Results The p16 and p53 expression levels were significantly decreased in the rats with mammary gland hyperplasia from Jianruling-treated groups, and lower than those in rats of model control group （p 〈 0.01 ）. Conclusion Jianruling can exert certain regulatory and therapeutic effects on mammary gland hyperplasia in rats.
