摘 要:目的:探讨炎性细胞因子IL-1α、L-1β、IL-6和TNF-α在尿结石形成中的作用,以及α-亚麻酸(α-LNA)对其的抑制影响。方法:将45只雄性Wistar成年大鼠随机分成3组,分别接受普通饲料加自来水饮水(A组)、普通饲料加诱石剂饮水(B组)和普通饲料加诱石剂饮水并每天用苏子油灌胃(2 g.只-1.d-1)(C组)。于喂养第8周后检测各组大鼠血细胞因子水平。利用体外培养的大鼠肾系膜细胞(RMC),设立空白对照(a组)、IL-6处理(b组)、EPA10μmol加IL-6处理(c组)、EPA50μmol加IL-6处理(d组)及无水乙醇处理(e组),用放免法检测培养上清中PGE2含量。结果:C组和A组IL-1αI、L-6水平显著低于B组,三组间IL-1β、TNF-α水平差异无统计学意义。细胞培养上清PGE2含量,a、c、d组显著低于b组,d组显著低于c组,a、e两组间差异亦无统计学意义。结论:炎性细胞因子IL-1αI、L-6可能参与了尿石形成过程,α-亚麻酸可能通过抑制炎性细胞炎子的产生而对尿石形成起保护作用。Objective:To investigate the possible role of cytokines interleukin (IL)-1β, IL-1α, IL-6 and TNFα in urinary stone formation and the inhibiting effect of α-Linolenic acid on these cytokines. Methods: Experiment 1 : 45 male adult wistar rats were divided into 3 groups: Group A was negative control, Group B was stone group (drinking 1% of ethylene glycol plus 1% of muriat water), Group C was perilla frutescens Britt · oil group (drinking 1% of ethylene glycol plus 1% of muriat water, meanwhile, tube feeding with perilla frutescens Britt · oil (10 g · kg^-1·d^-1). After 8 weeks, serum IL^-1β, IL-1α, IL-6 and TNFa were determined. Experiment 2: In vitro cultured RMCs were divided into 5 groups: Group a: control; Group b: pretreated with IL6; Group c: 10 μm EPA plus IL6; Group d: 50 μm EPA plus IL6 for 24 hours; Group e: pretreated with absolute alcohol. The PGE2 concentration in supernatants of RMCs was measured by radioimmunoassay. Results: Serum concentration of IL-1α, IL6 in group A and C rats were significantly lower comparing with those in B group rats. There weren't significantly differences among three groups for IL-1β and TNFα concentration. The PGE2 concentration in group a, c, d were significantly lower than that of group b. Group d was significantly lower than that of group c. There weren't difference between group a and e. Conclusions: Cytokines IL-1α and IL-6 may participate in urinary tone formation, αLinolenic acid may prevent urinary stone formation by the inhibitory role on these cytokines.
