摘  要:目的探讨中国北方汉族人群PLA2G4C基因多态性与精神分裂症的遗传相关性。方法采用聚合酶链反应(PCR)和限制性内切酶片段长度多态性(RFLP)方法,在240个精神分裂症患者核心家系中检测PLA2G4C基因上的6个单核苷酸多态性(SNPs),对结果进行单倍型相对风险(HRR)分析和传递不平衡分析(TDT)。结果在所检测的6个SNPs中,只有rs1366432和rs1549637位点在北方汉族人群中具有高度的多态性。rs1366432和rs1549637位点在精神分裂症病例组和对照组中基因型分布均符合Hardy-Weinberg平衡。HRR和TDT分析表明,rs1549637位点与精神分裂症关联具有显著性(χ2=5.364,P<0.05;χ2=5.633,P<0.05),而rs1366432位点与精神分裂症不存在遗传相关性(χ2=0.027,P>0.05;χ2=0.007,P>0.05)。结论PLA2G4C基因可能是精神分裂症的易感基因。Objective To investigate the genetic association between PLA2G4C gene polymorphism and schizophrenia in the North Han Chinese. Methods The method of PCR-based restriction fragment length polymorphism (PCR-RFLP) was applied to genotype 6 single nucleotide polymorphisms (SNPs) of PLA2G4C gene among 240 pedigrees consisting of fathers, mothers and affected offsprings with schizophrenia. Haplotype relative risk (HRR) test and transmission disequilibrium test (TDT) were conducted to analyze the genotyping data. Results In total of these 6 SNPs, rs1366432 and rs1549637 showed highly polymorphic in the North Han Chinese population. They were then applied as genetic markers to test the genetic association of PLA2G4C gene and schizophrenia. The genotypic frequency of PLA2G4C gene did not deviate from Hardy-Weinberg equilibrium in both case and control groups. HRR and TDT showed that rs1549637 was associated with the illness (X^2 = 5. 364, P = 0.021 ; X^2 = 5. 633, P = 0. 018). There was no association between rs1366432 and schizophrenia (X^2 = 0. 027, P = 0. 870 ; X^2 = 0. 007, P = 0. 936). Conclusions The PLA2G4C gene may be a susceptible gene of schizophrenia.
