核转录因子-κB在膀胱癌中作用机制的初步研究

摘  要:膀胱移行细胞癌石蜡包埋标本54例,正常对照标本13例。采用免疫组织化学SP法检测NF-κBP65和PKC蛋白在54例膀胱癌中的表达情况,运用TUNEL法原位检测凋亡细胞。分析NF-κB与PKC、细胞凋亡之间的关系,探讨NF-κB在膀胱癌中的可能作用机制。结果表明,NF-κBP65和PKC蛋白在54例膀胱癌标本中阳性表达率分别为:88.9%、81.5%,显著高于正常膀胱标本(P<0.01);54例膀胱癌中细胞凋亡的发生率为88.9%,凋亡指数均数为29.5741,均高于正常膀胱组织(P<0.01);NF-κBP65蛋白的表达与PKC蛋白的表达呈正相关(r=0.506,P<0.01),与细胞凋亡呈负相关(r=-0.390,P<0.01)。因此推论,NF-κB可能被PKC激活后,通过某种途径抑制细胞调亡,促进膀胱癌的发生和发展。In order to study the mechanism of pathogenesis of bladder carcinoma, the expression of NF-κBP65 and PKC protein was detected in 54 cases of forralin-affixed, paraffin-embedded bladder transitional cell carcinoma by immunohistochemical SP method,and TDT-mediated Deoxyuridine Triphosphate-biotin nick end labeling(TUNEL) method was also used to detect apoptotic tumor cells. 13 cases of normal bladder were assigned to controlled group. The correlation between NF-κB and PKC, NF-κB and cell apoptosis was analyzed, the possible mechanism of NF-κB in bladder carcinoma was explored. The results showed that the positive rates of NF-κBP65 and PKC protein in bladder transitional cell carcinoma were 88.9% and 81.5% respectively,and that in the controlled group were 7.7% and 7.7% respectively,the difference were significant ( X^2 = 31. 146, P 〈 0.01 ; X^2 = 22.630, P 〈 0.01 ). The rate of apoptotic bladder transitional cell carcinoma cells and the mean of AI were 88.9% and 29.5741 respectively,and in the controlled group were 15.4% and 0.6923 respectively, the difference were all significant ( X^2 = 26. 140, P 〈 0.01 ; t = 5. 496, P 〈 0.01 ). NF-κBP65 protein was positively correlated with PKC protein in bladder transitional cell carcinoma( r = 0. 506, P 〈 0.01 ), but was negtively associated with apoptotic tumor cells( r = - 0. 390, P 〈 0.01). It is deduced that NF-κB is possibly activated by PKC, then inhibit the cell apoptosis, and promote the occurrence and progression of bladder carcinoma.
