应用差别PCR技术研究胃癌中c-erbB-2原癌基因的扩增与胃癌的关系

摘　　要：目的探讨进展期胃癌组织中c-erbB-2原癌基因扩增情况及其与胃癌临床病理的关系。方法应用差别PCR技术检测47例进展期胃癌组织中c-erbB-2原癌基因的扩增。结果(1)胃癌中c-erbB-2原癌基因扩增率为34.0%(16/47);(2)胃癌侵及肌层c-erbB-2原癌基因扩增率为13.3%,侵及浆膜层组织c-erbB-2原癌基因扩增率为43.8%,两者有显著差异;(3)胃癌组织中c-erbB-2原癌基因扩增与患者年龄、性别、肿块位置、肿块大小、分化程度、淋巴结转移无显著相关性。结论c-erbB-2原癌基因扩增与胃癌的发生有一定关系,与进展期胃癌的浸润能力密切相关,在胃癌的诊断、治疗、预后判断中具有临床应用价值。Objective To explore the amplification of c - erbB - 2 oncogene in gastric carcinoma and its association with clinical pathology. Methods c- erbB- 2 oncogene amplification was detected by differental polymerase chain reaction in 47 cases of gastric carcinoma. Results （ 1 ） c - erbB - 2 amplification was found in 16 of 47 cases, the amplificated rate was 34.0%; （2）Significant association was shown between am- plification of c- erbB- 2 and the depth of tumor invasion; （3）c - erbB - 2 amplification was not associated with patients＇ age, gender, tumor size, location, differentiated type, lymph node metastasis. Conclusion The results suggest that c- erbB- 2 amplification play a role in the process of advanced gastric caricinoma and strongly suggest c - erbB - 2 amplification is associated with tumor＇ s invasion. This may provide a new approach for diagnose, therapy and predicting prognosis for gastric carcinoma.
