摘　　要：目的研究大黄酸衍生物RH-01对鼻咽癌KB细胞的抑制作用。方法体外培养鼻咽癌KB细胞,应用MTT法测定IC50,荧光染色观察细胞凋亡,流式细胞仪检测细胞凋亡率及细胞周期分布。结果RH-01明显抑制KB细胞的生长,半数抑制浓度(IC50)约为(0.67±0.20)μmol/L;0.8μmol/LRH-01作用24h即可明显诱导KB细胞的凋亡;流式细胞仪检测到细胞周期G2期阻滞明显。结论RH-01对人类鼻咽癌细胞有极强生长抑制和诱导凋亡作用。To study the effects of rhein derivative RH-01 on KB cells.Methods KB cells were treated with RH-01 at different concentrations. The growth inhibition was analyzed by MTr assay and mapping cell growth curves. Induced apoptesis was assessed by fluorescency microscopy. Cell cycle distribution was evaluated by flow cytometric analysis. Results RH- 01 remarkably inhibited the growth of KB cells,the IC50 value being（ 0.67 ± 0.20） μmol/L. Apopotesis of KB cells could be efficiently induced by RH-01 at concentration of 0.8 × 10^-6mol/L. Flow cytomatric analysis showed that RH- 01 could block cell cycle in the G2 phrase. Conclusion RH- 01 has potent cytotoxic effects on KB cells by inducing apoptesis and causing cell cycle arrest to G2/M phase in a dosedependent manner.
