摘 要:目的探讨腹腔注射卡介苗基因组DNA(BCGDNA)对哮喘小鼠模型的影响。方法BALB/c小鼠经鸡卵蛋白(OVA)免疫建立哮喘模型,分别于致敏前、激发时腹腔注射100μgBCGDNA,观察哮喘小鼠支气管肺泡灌洗液(BALF)中细胞总数和嗜酸性粒细胞数(EOS),ELISA检测脾单核细胞(SMNC)培养上清和BALF中白细胞介素4(IL4)、IL5、IL12及γ干扰素(IFNγ)的含量变化,常规病理切片观察肺内炎症情况,ABPAS染色观察杯状细胞增生和黏液分泌情况。结果哮喘小鼠OVA致敏前及激发时腹腔注射BCGDNA后可明显增加BALF和脾单核细胞培养上清中IL12和IFNγ的产生,使IL4和IL5的产生减少,BALF中嗜酸性粒细胞减少,黏液过度分泌明显减轻。结论哮喘小鼠腹腔注射BCGDNA可诱导IL12和IFNγ产生,抑制Th2型细胞反应,减轻哮喘气道炎症。Objective To investigate the eftects of intraperitoneally administered Mycobacterium bovis bacillus Cahnette Guerin( BCG)-DNA on a routine model of asthma. Methods The asthma model was established by immunizing BALB/c mice with ovalbumin(OVA). BCG-DNA (100μg) was intraperitoneally administered before OVA sensitization or OVA challenge.Total cell count and eosinophil (EOS) count in brochoalveolar lavage fluid (BALF) were measured. The levels of interleukin-4 (IL-4), IL-5, IL-12, and IFN-γ in splenic mononuelear cell( SMNC)culure supernatiant and broehoalveolar lavage fluid(BALF) were determined using Enzyme-Linked-hnmunn-Sorbent Assay (ELISA). Pulmonary inflammation was observed on routine pathological slides and the hyperplasia and hypcrsecretion of goblet cells stained with Alcian blue/periodic acid Schiff( AB- PAS)were also observed. Results BCG-DNA administered intraperitnneally before OVA sensitization and on OVA challenge significantly, inereased the levels of IL-12 and IFN-γ while decreased the levels of IL-4 and IL-5 in BALF and SMNC cuhtu'e supernataut. BCG-DNA administration also reduced the amount of eosinophils in BALF,suppressed mucus produetion and allergic' airway inflammation. Conclusions Intrapefitoneally administered BCG-DNA induces production of IL-12 aud IFN-γ and inhibites Th2 responses resulting a suppression of allergic airway inflammation. Our findings provide a new protocol for the treatment anti prevention of asthma.
