摘  要:目的从噬菌体随机肽库中初步筛选表达于SiSo细胞系的受体结合癌抗原(receptorbindingcancerantigenexpressedonSiSocells,RCAS1)的模拟表位。方法用抗RCAS1单克隆抗体2211对噬菌体随机肽库进行多级亲和筛选,建立与2211具有亲和力的肽库,随机挑选阳性克隆,用双抗体夹心ELISA和斑点ELISA检测其特异性。结果经过3轮免疫筛选,阳性噬菌体克隆富集了100倍,在随机挑取的10个阳性克隆斑中,有7个克隆可与2211单克隆抗体特异性结合。结论这7个阳性克隆中可能包含了肿瘤相关抗原RCAS1的模拟表位。Objective To identify the short peptides from a phage-displayed random peptide library through screening the epitopes of receptor-binding cancer antigen expressed on SiSo cells ( RCAS1 ). Methods MoAb 22-1-1 was used to immunoscreen a phage-displayed random peptide library. The positive clones were obtained by three rounds of biopanning, and the reactivity of each clone binding to MoAb 22-1-1 was examined by double-antibody sandwith ELISA and Dot ELISA. Results The eluted phages were enriched nearly 100 fold through three rounds of biopanning. 10 phage clones from the third round biopanning were randomly selected and seven of them could bind to the MoAb 22-1-1. Conclusion Epitopes of RCAS1 may be included in these seven positive clones.
