摘  要:目的探讨生长激素对胰腺癌细胞株Bxpc-3增殖活性的影响。方法MTT法检测生长激素对胰腺癌细胞株Bxpc-3增殖活性,Tunel法标记凋亡阳性细胞、免疫组化法标记增殖细胞,激光共聚焦显微镜计数免疫荧光双标Tunel阳性细胞和增殖细胞。结果10-7mol/L、10-8mol/L、10-9mol/L浓度的生长激素在第2天明显的抑制胰腺癌细胞生长,伴随作用时间的延长这种抑制作用逐渐减轻,且出现了加速胰腺癌细胞生长的作用,第4天、第5天胰腺癌细胞的生长同正常对照组相比有显著差异(P<0.01)。结论本实验研究从细胞学的角度证实生长激素在开始阶段具有抑制胰腺癌细胞生长的作用,但是这种作用伴随时间的延长逐渐消失,且出现加速了肿瘤细胞的生长的现象,为临床肿瘤病人应用生长激素提供了一定的理论依据。Objective To investigate the modulating effects of somatotropin on growth of human pancreatic cancer cell line Bxpc-3. Methods The Bxpc-3 pancreatic cancer cells were treated with somatotropin. The cell hyperplasia was detected by MTT and the apoptotic cells quantitatively determined by TUNEL. Then the proliferating cells and TUNEL-positive apoptotic cells were marked and determined by quantitative immune fluorescent technique. Results The growth of pancreatic cancer cells was limited by 10^-7mol/L, 10^-8mol/L and 10^9mol/L somatotropin on the 2nd d. The limiting effect began to decrease on the 3rd d. The cell proliferation was increased by somatotropin on the 4th and 5th d. Conclusions The pancreatic cancer cells are restrained by somatotropin at the beginning and then their growth is enhanced.
