摘 要:目的探讨人乳腺癌中原癌基因c-myc与hTERT mRNA表达的关系。方法收集癌旁正常乳腺组织、导管原位癌、浸润性导管癌各12、18和25例,用原位杂交法检测hTERT mRNA表达,并用免疫组织化学法检测乳腺导管癌c-myc表达。结果①hTERT mRNA在癌旁正常乳腺组织中未见表达,在导管原位癌、浸润癌中的阳性率分别为83.33%和88.00%,后两组hTERT mRNA表达明显高于正常组;②hTERT mRNA表达与浸润性导管癌肿块大小及淋巴结转移与否无相关性(P>0.05);③43例乳腺导管癌中hTERT mRNA与c-myc表达呈正相关(γ=0.3884,P<0.05)。结论端粒酶hTERT的表达可能在乳腺导管癌的组织发生中起关键作用,c-myc的过度表达可能与乳腺导管癌hTERT基因转录激活有关。Purpose To study the effect of protooncogene c-myc on the expression of hTERT mRNA in breast carcinoma. Methods The expression of hTERT mRNA was examined by in situ hybridization in 12 cases of normal breast tissue nearby cancer, 18 of ductal carcinoma in situ and 25 with invasive ductal carcinoma. The expression of c-myc was examined by immunohistochemistry in 43 cases of carcinoma. Results ① hTERT mRNA was not detected in normal breast tissue nearby cancer. The positive rate of hTERT mRNA in ductal carcinoma in situ and invasive ductal carcinoma were 83.33 % (15/18), 88 % (22/25), respectively. The prevalence and intensity of hTERT mRNA expression were much greater in carcinoma than those in normal breast tissue nearby cancer. ②The expression of hTERT mRNA was not correlated with tumor size and lymph node metastasis(P〉0.05). ③The positive correlation between hTERT mRNA and c-myc was found(γ=0.3884, P〈0.05). Conclusions hTERT mRNA expression is closely related to the malignant transformation of breast tissue. Overexpression of c-myc may play a role in the transcriptive activation of hTERT gene in breast carcinoma.
