中药PT液对衰老模型小鼠脑部凋亡细胞和Fas抗原的影响

摘　　要：目的 本实验是研究中药PT液对衰老模型小鼠脑部凋亡细胞和Fas抗原的影响。方法 昆明种雄性小白鼠40只随机分成4组，正常对照组、模型组、小剂量治疗组（PT1）和大剂量治疗组（PT2）：8用后，断头处死，冰上取鼠的脑部，制成石蜡切片，做凋亡细胞的表达和Fas抗原的免疫组化染色，随后计算凋亡细胞指数（AI）、Fas阳性细胞率和做Fas抗原的免疫图像分析。结果 模型组的AI和Fas阳性细胞率与对照组比较有显著性差异（P〈0．05）；PT1组的AI与对照组比较有显著性差异（P〈0．05），PT1组的Fas阳性细胞率和PT2组的AI、Fas阳性细胞率与对照组比较都无显著性差异（P〉0．05）；PT1组的AI和Fas阳性细胞率与模型组比较无显著性差异（P〉0．05）；PT2组的AI和Fas阳性细胞率与模型组比较有显著性差异（P〈0．05）。结论 PT液对治疗衰老具有较好疗效，Fas抗原较之AI对PT液更敏感：PT液可阻断衰老模型中由Fas抗原介导的凋亡启动程序。Objective To study Chinese medicine PT liquid how to affect the apoptosis cell and Fas antigen of senescing little rats＇ brain. Methods 40 Male little white rats of Kunming species are randomized into 4 groups, normal control group （control group）, model group, little dose treating group （PT1） and big dose treating group （PT2）; after 8 weeks , executed by decollation. Then take out rats brain on the ice , crank out paraffin slice and do the expression of apoptosis cell and immunity histochemistry dyeing of Fas antigen, followed by calculating apoptosis cell index （AI）, Fas masculine cell rate and doing immunity image analysis of Fas antigen. Results AI and Fas masculine cell rate of model group is significant discrepant with control group （P〈0.05）; AI of PT1 group is significant discrepant with control group （P〈0.05）, Fas masculine cell rate of PT1 group and AI, Fas masculine cell rate of PT2 group are not significant discrepant with control group （P〉 0.05）; AI and Fas masculine cell rate of PT1 group are not significant discrepant with model group （P〉 0.05）; AI and Fas masculine cell rate of PT2 group are significant discrepant with model group （P〈0.05）. Conclusion To treat senescing, PT liquid have good effect, Fas antigen is more sensitive than AI; PT liquid can interdict apoptosis start-up programme introduced by Fas antigen in senescing model.
