摘 要:目的:探讨表皮生长因子受体(epidermal growth factorr eceptor,EGFR)反义脱氧寡核苷酸硫代修饰片段对人肝癌细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)基因表达的影响。方法:应用逆转录聚合酶链反应(RTPCR)法,以次黄嘌呤磷酸核糖转移酶(hypoxanthine phosphoribosyl-transferase,HPRT)为内标,分析从1.6~3.2μmol/L浓度的EGFR反义脱氧寡核苷酸硫代修饰片段在84h时对人肝癌细胞VEGF基因mRNA表达水平的影响。结果:EGFR反义脱氧寡核苷酸硫代修饰片段有明显的抑制人肝癌细胞表达VEGF的作用,且这种抑制作用随着EGFR反义脱氧寡核苷酸硫代修饰片段浓度的增高而加强。结论:EGFR反义脱氧寡核苷酸硫代修饰片段能抑制人肝癌细胞VEGF基因的表达,为肝癌的抗血管治疗提供了新的途径和方法。Objective: To investigate the effect of phosphorothioate oligodeoxynucleotides (S- ODNs) of epidermal growth factor receptor (EGFR) on the regulation of vascular endothelial growth factor (VEGF) gene expression in human hepatocellular carcinoma cells. Methods: Treated with concentration range of 1.6-3.2μmol/L S-ODNs in culture medium,the relative VEGF mRNA level at 84th hour in human hepatoma cells was evaluated by reverse-transcriptase polymerase chain reaction (RT-PCR) with hypoxanthine phosphoribosyhransferase (HPRT) as an internal control standard. Result: The expression of VEGF gene mRNA was inhibited obviously by S-ODNs, and the inhibition was dependent upon the dose. Conclusion: SODNs can regulate and inhibit the expression of VEGF gene in human hepatocellular carcinoma cells, which might contribute to the development of a new therapeutic strategy for anti-angiogenesis.
