摘  要:目的研究脑出血大鼠延髓内脏带(MVZ)内孤束核、迷走神经背核FOS蛋白表达变化规律,探讨MVZ在脑源性多器官功能障碍综合征(MODS)的神经免疫调节作用.方法(1)向大鼠尾状核内注射Ⅶ型胶原酶0.8 U建立脑出血模型,54只Wistar大鼠随机被分为对照组、假手术组及出血后4、8、12、24、36、48和72 h时相点的7个亚组;(2)应用免疫组织化学ABC法观察各时相点MVZ内孤束核、迷走神经背核FOS蛋白的表达变化规律.结果(1)在对照组和假手术组MVZ内孤束核、迷走神经背核FOS蛋白阳性表达极少;出血组FOS蛋白阳性细胞在孤束核、迷走神经背核密集表达,24~36 h达高峰,在72 h仍有阳性表达.结论孤束核、迷走神经背核FOS蛋白的阳性表达增多提示MVZ是脑出血致MODS的神经免疫调节中枢之一.Objective To investigate the changes of FOS protein expression in the nucleus of the solitary and dorsal nucleus of vagus nerve of medullary visceral zone (MVZ) following cereral hemorrhage and to discuss the possible center regulation mechanism of MVZ in the cerebrogenic multiple organ dysfunction syndrome (MODS). Methods (1) Cerebral hemorrhage was induced in rats by injecting 0. 8 U collagenase Ⅶ into the caudase putamen. 54 Wistar rats were randomly divided into 9 groups:normal group, sham-operative group and hemorrhage groups including 4th h,8th h, 12th h,24th h,36th h,48th h and 72 th h in seven time points. (2) The expression of FOS protein in the nucleus of the solitary and dorsal nucleus of vagus nerve of MVZ were measured by immunohistochemistry (ABC). Results A few of FOS protein expressd in the nucleus of the solitary and dorsal nucleus of vagus nerve of MVZ of normal control group. The FOS-like neurons densely expressed in the nucleus of the solitary and dorsal nucleu of vagus nerve of cerebral hemorrhage group, FOS protein expresses in the MVZ with time progress reaching the peak at the 24-36 h, FOS-like neurons can be observed at the 72 h still. Conclusions FOS protein expressed in the nucleus of the solitary and dorsal nucleus of vagus nerve may contribute to dysfunction of peripheral organs. MVZ may be one of the regulation center after cerebral hemorrhage by MODS.
