摘　　要：目的阐明转录共激活因子(p300)在大鼠组织中的分布特点及其表达的组织差异,探讨p300在组织差异相关的基因表达调控中的作用。方法采用免疫组织化学技术检测p300在雄性大鼠大脑皮质、海马、小脑、心肌、肝脏、肺脏、肾脏、睾丸中的分布特点;运用免疫印迹技术,对不同组织中p300蛋白水平进行检测。结果①p300蛋白在心肌、肝脏、肺脏、肾脏、睾丸、小脑、海马、大脑皮质中均有分布。②在睾丸、海马和肾脏组织中,p300主要分布在精原细胞、锥形神经元细胞和远曲小管上皮细胞中;大脑皮质神经元均表达p300,在分子层中呈弱阳性;p300在小脑蒲肯野细胞和心脏组织的心肌细胞表达呈强阳性。p300在肝细胞中均匀分布,而肺脏组织中p300表达呈弱阳性。③p300蛋白在骨骼肌中表达水平最高,其次是心肌、睾丸、肝脏和小脑,下丘脑中未检测到明显的阳性表达。结论p300的表达和分布具有组织和细胞差异性,这种差异分布可能是机体不同组织、细胞中蛋白表达差异的重要机制。Objective To explore the distribution and expression pattern of the coactivator p300 in rat tissues. Methods The location of p300 protein in rat brain, heart, testis, liver, lung, kidney and skeletal muscle was detected by immunohistochemistry. The level of p300 protein in various tissues was detected by Western blotting. Results p300 was widely expressed in all the detected tissues. In cerebral cortex, cerebellum and kidney, p300 located mainly in the neurons, Purkinje cells and the epithelium cells of medulla respectively. p300 was expressed exclusively in the spermatogonium of testis and the pyramidal cells of hippocampus. The protein level of p300 was highest in the skeletal muscle, next in the heart, live testis and cerebellum. No strongly positive expression was detected in the hypothalamus. Conclusion The p300 protein expression is universal in rat tissues, but the location and expression are tissue- and cell-specific, which suggests that p300 may contribute to the protein expression difference in various tissues and cells of rat.
