摘 要:目的探讨PTTG的表达在非小细胞肺癌(NSCLC)发生、发展中的作用及其与PTEN和Ki67蛋白表达的关系。方法应用免疫组化SP法检测PTTG、PTEN和Ki67三种蛋白在60例NSCLC和19例正常支气管黏膜上皮组织中的表达。结果NSCLC组织中PTTG蛋白表达的阳性率为73.33%(44/60),显著高于正常支气管黏膜上皮组织中的表达(P=0.000);PTTG的表达与肿瘤细胞的分化程度和有无淋巴结转移显著相关,但与患者的年龄、性别及肿瘤的组织学类型无关;NSCLC组织中PTTG表达与PTEN表达无相关关系,而与Ki67表达呈显著正相关。结论NSCLC中PTTG高表达,PTEN低表达,PTTG和Ki67在NSCLC的发生和发展中有协同作用,三者可作为反映NSCLC生物学行为的指标。PTTG将有望成为新的肿瘤分子标记与基因治疗的靶点。Objective To investigate the expression of PTTG and its relationship with expressions of PTEN and Ki67 proteins in human non - small cell lung cancer( NSCLC). Methods Immunohistochemical methods were applied to detect the expression of PTTG, PTEN, Ki67 proteins in 60 surgical specimens from NSCLC patients and 19 normal bronchial epithelium. Results In NSCLC, the positive rate of PTTG protein were 73.33 % (44/60). Expression of PTTG protein were detected in a significandy greater proportion in NSCLC than that in normal bronchial epithelium, P = 0. 000 ; There was no relationship between PTTG expression and age,sex and histological subtype, respectively; otherwise, there was closely relationship between PTTG expression and cellular differentiation and lymph node metastases, respectively;Expression of PTTG was not correlated with expression of PTEN; Expression of PTTG was positively correlated with expression of Ki67. Conclusion Overexpression of PTTG and lower - expression of PTEN may be related to human NSCLC, PTTG and Ki67 play a cooperative role in the process of NSCLC, all of them may be used as important indices for biologic behavior of NSCLC. PTTG may be identified as new therapeutic targets.
