摘  要:目的研究67Ga ECT肿瘤显像评价低功率超声辐射微泡治疗小鼠肝癌腹水转移皮下瘤的生物学效应。方法KM昆明种小鼠6只建立小鼠H22腹水转移皮下瘤模型,随机分为A、B、C三组。A组单纯低功率超声治疗肿瘤;B组微泡剂+低功率超声治疗肿瘤。C组正常对照组,观察三组动物抗瘤效应、组织学、免疫组化检测PCNA、CD34蛋白表达、67Ga ECT肿瘤显像。结果三组小鼠肿瘤处理前后体积变化及生长率比较,B组肿瘤治疗后体积明显被抑制及生长率下降。A、C二组比较t=0.91,P>0.05;B、C二组比较t=2.95,P<0.05。形态学改变B组可见明显的肿瘤血管栓塞及肿瘤坏死。免疫组化检测PCNA、CD34在C组、A组呈强阳性表达,而B组表达明显下降。A、B、C三组PCNA阳性指数(PI)分别为89.9%、37.4%、91.7%;CD34MVD计数分别为78.4%、31.2%、77.1%。67Ga ECT肿瘤显像B组微泡剂+低功率超声治疗前后,除肝脏部位放射性明显浓聚外,肝癌皮下瘤小鼠右侧前肢见明显放射性浓聚,用ROI技术计算比较,治疗后肿瘤部位放射性明显降低,下降46.2%。结论低功率超声辐射微泡治疗小鼠肝癌腹水转移皮下瘤,可以致肿瘤血管栓塞及抑制肿瘤细胞的增殖;67Ga ECT肿瘤显像评价低功率超声辐射微泡致肿瘤血管栓塞有一定作用。Objectives To study the biological effect of the low power ultrasound irradiation treatment to the transplanted hepatic cancer model in mice by ECT imaging. Methods Six hepatic carcinoma model mice were randomly divided into three groups (group A.. low power ultrasound irrradiation treatment; group B; microbubble-CO2 +low-power ultrasound irradiation treatment; group C: control). The hepatic cancer model was established in mice via the endermic tumor transplantation of transfer hepatic carcinoma ascites. The tumor resistant effect, pathological histology, proliferating cell nuclear antigen (PCNA) and CD34 of immunohistochemistry, and tumor positive imaging by ^67Ga were observed and analyzed. Resalts The volume and the growth rate of tumor bodies were compared among three groups of mice. The volume and the growth rate of tumor body decreased distinctly in group B. (Group A and group C: t= 0.91, P〉0.05 ; Group B and group C.. t=2.95, P〈0.05). The pathological histology observed that there was micro-vessel embolization and necrosis area in the center of tumor in group B. Immunohistochemistry analysis that PCNA and CD34 were strongly positive in both group A and C and they were decreased distinctly in group B. Among three groups of mice, PCNA proliferating index (PI) was 89.9%, 37.4%, 91.7% respectively and CD34 microvessel density (MVD) was 78.4%, 31.2%, 77. 1% respectively. The tumor positive imaging by ^67Ga show that in group B there was marked radioactivity concentration in the right frontal limb area besides in the liver area pre-and post-treatment, but the radioactivity counts was decreased distinctly in tumor body after treatment through ROI calculation, decreasing 46. 2%. Conclusion The hepatic carcinoma model mice were treated by the irradiated microbubble-CO2 of low-power ultrasound, inducing micro-vessel embolization of tumor body and restraining reproduction of tumor cells.
