摘  要:目的:检测caspase-3基因及其底物多聚ADP-核糖聚合酶(PARP)在人肺癌组织的表达及与临床病理特征的关系。方法:采用第2代免疫组化Elivision法和Western blot法检测57例人肺癌原发灶中caspase-3、PARP蛋白的表达。结论:通过Western blot和免疫组化检测caspase-3、PARP的蛋白表达结果一致,在57例肺癌原发灶中caspase-3、PARP 2种蛋白表达阳性率分别为:66.67%,33.33%。caspase-3与细胞分化程度(P=0.023)、淋巴结转移(P=0.004)和组织学类型显著相关(P=0.036),PARP与淋巴结转移显著相关(P=0.025)。57例人肺癌组织中,caspase-3的表达与PARP呈显著负相关(P=0.001)。结果:caspase-3通过降低其底物PARP的表达从而促进肿瘤细胞的凋亡,明显地抑制了肺癌的侵袭和转移。Objective: To investigate the relationship between the expressions of caspase-3 gene, and its substrates Poly ADP-ribose polymerase (PARP) and the clinicalpathological characteristic of pulmonary carcinoma. Methods: Caspase-3 and PARP protein levels in 57 patients with pulmonary carcinoma (21 patients with squamous cell carcinoma, 27 with adenocarcinoma, 4 with large cell carcinoma and 5 with small cell lung carcinoma) were detected by the second generation immunohistochemical Elivision methods and Western blot. Results: In 57 pulmonary carcinomas, the coincident results of caspase-3 and PARP proteins were observed by Western blot and immunohistochemistry, caspase-3and PARP proteins positive rates were 66.67%, 33.33% respectively. The positive expression of caspase-3 protein was highly associated with cellular differentiation degree (P=0.025), lymph node metastasis(P=0.004) and the pathological types(P=0.036). PARP expression was only associated with lymph node metastasis(P=0. 023). In 57 pulmonary carcinomas, a negative correlation was present between caspase-3 and PARP protein expression (P = 0. 001). Conclusion: Caspase-3 obviously inhibits invasion and metastasis of pulmonary carcinoma by extensively degrading target prc)teins such as PARP.
