摘  要:目的:观察Cox-2抑制剂(塞来昔布、罗非昔布)对人骨肉瘤细胞株HOS-8603、乳腺癌细胞株MDA的作用。方法:采用MTT比色法测定体外培养的细胞株HOS-8603、MDA-MB-435S对不同浓度的罗非昔布和塞来昔布的敏感性。流式细胞仪分析细胞增殖周期的影响。结果:选择性环氧化酶-2(Cox-2)抑制剂明显抑制HOS-8603、MDA-MB-435S的生长(P<0.05),抑制效应呈现浓度依赖型和时间依赖型,HOS-8603在Cox-2抑制剂作用下,细胞分裂阻滞在G2/M期,有一定的诱导细胞发生凋亡作用,塞来昔布强于罗非昔布对HOS-8603的作用。结论:选择性环氧化酶-2抑制剂对骨肉瘤细胞、乳腺癌细胞的生长可能起着十分重要的作用,有望成为成骨肉瘤治疗和抑制骨肉瘤转移的新药物。Objective: To compare the effects of two kinds of selective cyclooxygenase-2 inhibitors (celecoxib, rofecoxib) on the growth of human osteosarcoma and breast cancer cell line. Methods: The anti-proliferative effect was measured by MTT assay. Cell cycle and apoptosis were analyzed by flow cytometry. Results: Cyclooxygenase-2 specific inhibitor had a growth inhibitory and apoptosis-inducting effect on three kinds of tumor cell. The inhibition effects showed a dose-dependence and time-dependence manner. MTT assay indicated a significant difference between the rofecoxib and celecoxib group. The cells treated with Cyclooxygenase-2 specific inhibitors showed G2/M arrest. Moreover, celecoxib was more inhibitorily higher than rofecoxib. Conclusion: Cyclooxygenase-2 specific inhibitors have significant effect on the growth of two kinds of cell line. Cyclooxygenase-2 specific inhibitors may become one of the important medicines in the treatment and metastasis of tumor.
