新城疫病毒疫苗对人卵巢癌细胞株治疗作用的研究

摘　　要：目的:研究新城疫病毒疫苗(ATV-NDV)对人卵巢癌细胞株体外生长的抑制作用。方法:制备人卵巢癌细胞株3AOATV-NDV瘤苗,将不同浓度的ATV-NDV、白细胞介素12(IL-12)、白细胞介素15(IL-15)、ATV-NDV+IL-12、ATV-NDV+IL-15分别与正常人外周血淋巴细胞混合培养,MTT法测定其刺激淋巴细胞增殖的程度及对肿瘤细胞的生长抑制率;HE染色法观察效靶比为50∶1时活化的淋巴细胞与3AO细胞共同培养24～96h后细胞形态学的变化。结果:活化的淋巴细胞与靶细胞共培养可杀死肿瘤细胞,并具有时效性(P<0.05);不同细胞间细胞生长抑制率(GIR)差异无显著性(P>0.05);IL-15、IL-12与ATV-NDV在活化淋巴细胞方面具有协同作用,与单独作用差异有显著性(P<0.01);IL-15与ATV-NDV作用差异无显著性(P>0.05)。淋巴细胞作用48h后3AO细胞即出现凋亡形态。结论:经ATV-NDV、IL-12、IL-15、ATV-NDV+IL-12、ATV-NDV+IL-15混合培养的正常人外周血淋巴细胞可有效地抑制3AO细胞株的生长,并具有时间、浓度依赖性,其作用机制是诱导细胞凋亡。Objective：To investigate the inhibiting effect of autologous tumor cell vaccines-chicken Newcastle disease virus-infected （ATV-NDV） on the growth of different human ovarian carcinoma cell strains in vitro. Methods： Normal human ovarian epithelium cells and human ovarian carcinoma cell strain SKOV3 were cultured in vitro. Peripheral blood lymphocytes were separated from normal individual and autologous tumor cells vaccines-ATV-NDV with human ovarian carcinoma cell strain 3AO were prepared. ATV-NDV,IL-12,IL-15,ATV-NDV＋IL-12 and ATV-NDV＋IL-15, in different concentration and action time, were respectively cultured with human peripheral blood lymphocytes. MTT assay was used to analyze the effect of cash reagent on the proliferation degree of lymphocyte and the cell growth inhibiting rate; hematoxylin-eosin （HE） staining was taken in order to observe the change of cell modality cultured for 24-96 hours with the E：T of 50：1. Results： The activated lymphocytes could kill carcinoma cells if they were cultured together with target cells,and the effect was promoted as the time prolonged （P〈0.05）.The difference of cell growth inhibitory rate （GIR） among different cells was not significant（P〉0.05）. IL-15,IL-12 had synergism with ATV-NDV on the effect of activating lymphocytes, and the difference was significant comparing with each alone （P〈0.01）. There was no difference between the effect of IL-15 and ATV-NDV （P〉0.05）. As the action time of lymphocyte prolonged,the 3AO cells faded and displayed the modality of apoptosis cells when lymphocytes had done for 48 hours, and necrosis cells formed when been done for 96 hours. Conclusion：ATV-NDV, IL-12 and IL-15 have effect on the proliferation of normal human peripheral blood lymphocytes and the human peripheral blood lymphocytes after cultured together with ATV-NDV, IL-12, IL-15, ATV-NDV＋IL-12 and NDV＋IL-15 can effectively inhibit the growth of the ovarian carcinoma cell strain 3AO, which has a time and concentration dependenc
