摘 要:目的:探讨组织多肽特异性抗原(TPS)、癌胚抗原(CEA)表达对肺癌细胞粘弹特性的影响,力图从生物力学角度揭示肺癌细胞侵润转移的某些生物学特性。材料与方法:体外培养的低转移人肺腺癌(PAa)细胞和高转移人肺巨细胞癌(PG)细胞经TPS、CEA检测试剂孵育后,应用酶联免疫吸附试验检测其孵育液中TPS、CEA表达水平,并采用微管吸吮技术定量测定癌细胞粘弹特性。结果:PG细胞的TPS表达高于PAa细胞,而PAa细胞的CEA表达高于PG细胞。肺癌细胞粘弹性,在未使用TPS、CEA检测试剂孵育组,为PAa细胞大于PG细胞;在TPS检测试剂孵育组,PA a、PG细胞粘性系数降低,弹性系数K 1,K2增高;在CEA检测试剂孵育组,PA a、PG细胞仅弹性系数增高。结论:TPS、CEA为肺癌的重要肿瘤标志物,TPS、CEA表达与PAa、PG细胞的粘弹特性有关,TPS表达与PG细胞的高转移性能有关。Objectives: To study the effect of expression levels of tissue polypeptide specific antigen (TPS)and eareinoembryonic antigen(CEA) for the viscoelastic properties of lung cancer cells. The authors try to reveal the relevant biomeehanieal and biorheologieul manifestations of human lung cancer. Materials and methods: The levels of TPS or CEA were determined by enzyme-linked immunmosorbent assay in the hatching fluids of the high metastatic human giant cell carcinoma (PG) cells as well as the low metastatic adenoeareinoma (PAa) cells of lung based on cell culture in vitro and hatched by the detection reagant of TPS or CEA. Using mieropipette aspiration technique, authors investigated the viscoelastic properties of lung cancer cells quantitatively. Viscoelasticity was analyzed by using the special three-element standard linear solid model. Results: No detection reagant of TPS or CEA, the elastic coefficients K1 and K2, as well as viscous coefficient μ of PAa cells were bigger than that of PG cells.Treatment with the detection reagant of TPS, the values of K1,K2 of PAa and PG ceils increased,and their values of μ decreased, Treatment with the detection reagant of CEA, the values of K1,K2 of PAa and PG cells increased only. Conclusions: TPS and CEA were important tumor markers of lung cancer. The expression levels of TPS or CEA were interrelated with the viscoelasticity of PAa and PG cells of lung; The expression levels of TPS were interrelated with the high metastatic properties of PG cells of lung.
