摘  要:以壳聚糖为原料,将其溶于稀酸,确定了碱性介质固化,洗涤至中性,再在中性介质中交联的工艺路线.对壳聚糖的脱乙酰度和质量浓度、交联剂戊二醛的用量、交联时间,以及反应介质等工艺条件进行了优化,最适工艺条件为:壳聚糖的脱乙酰度为90.24%,质量分数为2.0%;交联剂戊二醛质量分数为0.6%;交联时间为6 h;以水为反应介质.在此工艺条件下制备的凝胶在pH=1.5的介质中稳定.应用试验表明,对蛋白质有良好的分离效果,中性蛋白酶经一次分离,可得到四个组分,总活力保持在90%以上.牛血清蛋白经凝胶可分离出两个组分,蛋白回收率达70%以上.The chitosan was solid in the alkali phase tirstly, wasnea untu me neutral cross-linked in the neutral phase. Some technical conditions were optimized such as the alkali content of soliding the chitosan, the amount of cross-linking solvent glutaraldehyde, the cross-linking time, the degree of deacetylation of chitosan, the chitosan content, reaction phase and stirring form etc. The most optimization conditions: the degree of deacetylation of chitosan 90.24%, the chitosan content 2.0% ; the amount of cross-linking solvent glutaraldehyde 0.6 % ; the cross-linking time 6 h; water phase. A novel kind of gel was prepared under the optimized conditions. The protein could be separated well through the gel. Neutral protease was gotten four ingredients through the gel column, the total activity of which kept above 90%. Albumin Bovine Serum (BSA) could be gotten two ingredients through the gel column, the total yield of which reached above 70%.
