摘 要:目的通过研究双歧杆菌脂磷壁酸(lipoteichoic acid,LTA)对结肠癌细胞中Toll样受体(TLR)表达的影响,探讨TLR在启动双歧杆菌LTA抗肿瘤信号转导通路中的作用。方法用RT-PCR检测结肠癌细胞HCT-116经双歧杆菌LTA处理前后TLR的表达变化。结果结肠癌HCT-116细胞有TLR的基础表达,且经LTA处理后受体的表达增强,呈剂量依赖性。结论双歧杆菌LTA可上调TLR的表达,提示双歧杆菌LTA诱导肿瘤细胞凋亡的信号转导途径可能由TLR启动。Objective To explore the signal transduction pathway of lipoteichoic acid (LTA) of Bifutobacterium in anti-tumor mechanism through investigating the effect of LTA on the expression of Toll-like receptor(TLR) in colon cancer cells. Methods Changes of TLR expression in colon cancer cells HCT-116 treated with LTA was detected by RT-PCR. Results The basic expression of TLR in HCT-116 cells was confirmed, and after treatment with different dose of LTA of Bifutobacterium, the expressions of TLR( especially TLR2) were markedly increased in a dose-dependent manner. Conclusion LTA of Bifidobacterium can upregulate the expression of TLR in HCT-116 cells, it seems that TLR initiates the anti-tumor signal transduction of LTA of Bifidobacterium on apoptosis of colon tumor cells.
