摘 要:利用BSA法,以显性多子房小麦(Triticum aestivum)与普通小麦(T.aestivum)品种西农1376进行杂交,从F2分离世代建立多子房性状基因池和单子房基因池,采用RAPD-PCR方法,以440个随机引物筛选单、多子房的混合群体及其双亲。发现10个引物的扩增产物在近等基因系间表现出特异性,表现为多子房亲本及其混合群体有带,单子房亲本及其混合群体无带;6个引物扩增产物表现出的特异性相反,单子房有带,多子房无带。经过重复实验和群体单株验证,引物S236在多子房亲本、混合群体及其群体单株中都表现出稳定的多态性,可以作为多子房显性基因的分子标记。Using BSA method, a dominant multi-ovary line of common wheat (Triticum aestivum) was crossed with variety Xinong 1376 (T. aestivum). From F2 segregation population, a multi-ovary and a single-ovary gene pools were established; via RAPD-PCR method, single-ovary and multi-ovary mixed populations were screened using 440 random primers. The result indicated that the expanded product of 10 primers showed specific character among neaisogenic lines, multi-ovary population and its parent had band, while single-ovary population and its parent didn't. The expanded product of 6 primers showed the opposite character that the product of single-ovary line had band and that of multi-ovary line had not. After repeating tests and checking up individual plant, only primer S236 showed the stable multi-morphological character to multi-ovary parent, mixed population and single plant, so it could be used as a molecule marker of the dominant multi- ovary gene.
